基于肾脏足细胞自噬探讨苏木提取物对
Heymann 肾炎模型小鼠的肾保护作用

摘 要 目的:研究苏木提取物对被动型 Heymann 肾炎模型小鼠的影响及对足细胞自噬反应的作用机 制。方法:将实
验动物随机分成 5 组,即正常对照组.Heymann 肾炎模型组和低、中、高剂量苏木提取物 组,给药 21 d 后检测各组
小鼠 24 h 尿蛋白、血清白蛋白、肌肝等指标;光镜观察肾组织病理变化,免疫荧 光染色法检测肾脏 C5b-9 沉积,
蛋白印迹法(Western blot, WB)检测相关蛋白表达。结果:(1 ) 与正常对 照组相比,模型组 24 h 尿蛋白、血肌酹均不
同程度升高,血清白蛋白不同程度降低(P<0. 05); 模型组光 镜下可见肾组织明显分布不均匀,肾小球明显增大,系
膜区显著增宽;模型组肾小球毛细血管祥 C5B-9 沉积明显增强;模型组肾组织 P62 表达明显降低(P<0. 05),
ATG5、LC3 H 表达均明显升高(P<0. 05)

(2) 与模型组相比,苏木提取物各剂量组 24 h 尿蛋白、肌 Sf 均不同程度降低,血清白蛋白不同程度升高(P <0.
05);肾组织排列较不规范,部分肾小球出现增大;肾小球毛细血管#C5b-9 的沉积有所减少,肾组 织 P62 表达明显
升高,ATG5、LC3U 表达均明显降低(P<0.05)。结论:苏木提取物对 Heymann 肾炎小鼠 具有肾保护作用,通过调
控自噬进而保护和修复足细胞是其部分作用机制。

Discussion on Renal Protective Effect of Sumu (Hematoxylon) Extract on
Heymann Nephritis Model Mice based on Podocyte Autophagy

ABSTRACT Objective: To investigate the effect of Hematoxylon Extract on passive I leymann nephritis model mice
and the mechanism of podocyte autophagy reaction. Methods: The experimental animals were randomly divided into 5
groups: normal control group, Heymann nephritis model group and low, medium, high - dose He- matoxylon Extract
group. After 21 days of medication, 24 h urinaiy protein, serum albumin, creatinine and other indexes were detected in
each group. The pathological changes of renal tissues were observed by light microscopy. Immunofluorescence staining
was used to detect the deposition of C5B - 9 in the kidney. Western blot ( WB) was used to delect the related proteins
expressions. Results: (1) Compared with normal control group, 24 h urinary protein and serum creatinine in mode]
group were increased to vaiying degrees, while serum albumin was decreased to vaiying degrees ( P <0. 05) .
Compared with the normal group, the distribution of renal tissue in the model group was significantly uneven, the
gloniemlus was significantly enlarged, and the mesangial area was sig- nificcintly widened. The deposition of C5b - 9
in glomerular capillary loop was significantly enhanced in model group. The expression of P62 in renal tissue of model
group was significantly decreased (P < 0. 05) , while the expressions of Atg5 and LC3 were significantly increased (P <
0. 05). (2) Compared with model group, 24 h urinaiy protein and creatinine in Hematoxylon Extract groups were
decreased in different degrees, while serum albumin was increased in different degrees (P < 0. 05 ). the renal tissue
